Protein PKCI/HINT1 earlier already connected with a schizophrenia and bipolar affective frustration. At mice, for example, at removal of a corresponding gene characteristic reaction on декстроамфетамин (a stimulator of the central nervous system) which it was considered to be a schizophrenia symptom developed.
Authors have spent a number of tests in hope to establish connection between absence PKCI/HINT1 and any changes in behaviour of mice. Experimental techniques corresponded to the standard; mice started in a so-called water labyrinth of Morrisa, suspended for tails, forced to be tested compulsory swimming (the test of Porsolta).
The water labyrinth of Morrisa in this case represented round pool in diameter of 122 sm and depth of 25 sm, filled with an opaque mix of water and milk. In this pool on depth of 0,5 sm the round platform in diameter of 9 sm settled down, which mouse should find to get out outside; the animal could be guided in the subjects located around. As it is informed, mice with remote gene PKCI/HINT1 have shown in this experiment бóльшие abilities to training, than "usual" individuals, and after the several spent tests found a platform much faster.
In the test for compulsory swimming of rodents placed in the five-litre capacity filled with water of which they could not get out. Such test is often used for an estimation of action of energizers on animals; it is considered that the total of time spent by the mouse in an immovability, corresponds with its mood (in other words, the long immovability expresses despair). Similar criteria are established for experiment in which mice suspended for a tail: малоактивное the behaviour corresponds to bad "mood". According to authors, in both tests the mice deprived PKCI/HINT1, have proved to be more persevering and active.
The same results have been received and in following testing: rodents started in a cage divided into two compartments identical on the area one of which was shined, and another — is not present. It is known that mice prefer to be in a dark premise, however completely to refuse research of surrounding space cannot and all the same come into light branch. Thus animals with absent gene PKCI/HINT1 spent on light of much more time.
«Unfortunately, we are not ready to tell yet why loss of one gene influences mood of mice, — makes comments on Tszja Bej Van (Jia Bei Wang), one of researchers. — however already it is now clear that influence on protein PKCI/HINT1 can become one of components of an original technique of treatment of depression, a schizophrenia and bipolar affective frustration. Besides, the mice deprived PKCI/HINT1, can be used as models for studying of a maniacal syndrome».
